Introduction: Proteolysis-targeting chimera (PROTAC) estrogen receptor degraders are a type of hormone therapy. They break down estrogen receptors found on hormone receptor-positive cancer cells.

The PROTAC estrogen receptor degrader vepdegestrant (Veppanu) is used to treat hormone receptor-positive, HER2-negative metastatic breast cancers that have an ESR1 tumor gene mutation and have been treated with hormone therapy and a CDK4/6 inhibitor in the metastatic setting.

The ESR1 mutation is in the genes of the breast cancer cells, not the genes of the person. The American Society of Clinical Oncology (ASCO) and the National Comprehensive Cancer Network (NCCN) recommend all hormone receptor-positive, HER2-negative metastatic breast cancers be tested for ESR1 tumor gene mutations [1-2].
